Abstract

The 61-adrenergic receptor (gl All) mediales several tunction of tale cholamines such as stimulation of heart rate and cent reactivity. Sexat aldora ai us in lhe BI-AR promoter were identified by transiently translk, ctiR chin:eric genes containing the t:1-AI promoter driving luciIbrase inlo cells aim by lXa.se 1 footprinting and mobility-shift assays. A glucocorticoid responsive domain was localized between -950 and -926 relative to lhe iniliator Al. A multi functional region between -125 and 100 contained suppressiw, and hormone responsive domains. region between -121 and -118 diminished tho activity of the gene in the context of he 3 kb promoter. The sequence between -117 and -101 contained a thyroid hormone responsive element tha aclivatod B1 A R transcription in ventticular myocytes in response to T:l. Moreover. these sequences increased 61-AR transcription in F9 cells in respollSO to difforenti ation bv retinoie acid. The basal promoter was localized between -3S0 and -321. This region contained functional Spl and AP 2 and immediate early genes binding sites.
