Retinal bone marrow derived perivascular dendritic cells may present retinal antigens in induction of EAU

Abstract

Purpose. To address the issue of local antigen presentation in experimental autoimmune uveitis (EAU), we previously conducted in vivo studies m bone marrow chimeras and demonstrated the presence of bone marrow derived cells in the retina In the present study, expression of la molecules by these bone marrow derived retinal dendritic cells was investigated in chimenc rats following adoptive transfer of S-antigen specific T-cells Methods. Twenu one bone marrow chimeras [(Lewis x Brown Norway)F|-»Brown Norway] u ere constructed by transplanting 50 x 106 bone marrow cells of LBNF- (RT-lln) rats to lethally irradiated Brown Norway (RT-In) rats. Following adoptive transfer of 20 x 10s S-anti gen-specific, Lewis CD4 + T-lymphocytes, one group of three rats llas sacrificed each day for seven days Whole mount preparations of the retinae from the sacrificed rats were stained for Lewis-specific la (OX3), ICAM-1 and VCAM-1 molecules Similar studies were conducted in naive chimeras (three) and chimeras injected intravitreally with IFN-y (three) Results, la expression on retinal perivascular cells with dendritic morphology was seen on day 4 after adoptive transfer Retinal vascular endothelium was positive for ICAM-i from day I. however EAU was observed only after day 5 Expression of ICAM-1 and VCAM-1 llas seen in eyes of naive as well as IFN-y injected rats, whereas la expression was observed in eyes injected with IFN-y Conclusion. Although ICAM-1 was expressed on vascular endothelium, inflammation did not begin until after the expression of la molecules by the retinal perivascular dendritic cells Hence these dendritic cells may play a role in the induction of EAU and retinal perivascular distribution of inflammatory cells.
