Serological characterization of the globulin fractions of pigeonpea seed storage proteins

Abstract

Seed storage globulin fractions (legumin, vicilin and convicilin) of pigeonpea var. Pant A-3 were characterized serologically with respect to their immunogenicity, cross-reactivity and standardization of ELISA technique for their minute detection. Globulin fractions-legumin, vicilin and convicilin were obtained by zonal isoelectric precipitation from the total globulins. Antibody against two fractions namely vicilin and legumin were raised in New Zealand white rabbits and a titre value of 2560 and 1280 were obtained in the antisera respectively. Ouchterlony's double diffusion tests showed the cross-reactivity with the corresponding antigens but anti-vicilin was found to be cross-reactive to convicilin also. ELISA procedures were standardized to detect and quantify minute quantity of vicilin and legumin directly from any sample.
